Step 1:第一步

洗净鱼,用镊子在鱼皮下取下大米粒大小的鱼肉,注意不要让手指接触到样品。
   

Step 2:第二步

把鱼的样品放入含有清液的小瓶里。注意:手上的汗水、油脂会污染样品。

 

Step 3:第三步
从长管中拿出一个试剂条放入含清液和与样品的小瓶中,纸面的那一头朝下,等20分钟。

 

 

 

 

Step 4:第四步
用清水洗净镊子放回原处。拿出试剂条,并在空气中风干15-20分钟。手指等不要碰到纸面的那一头(白色的)。

 

 

 

 

Step 5:第五步
轻轻摇动小瓶,混合蓝色液体。打开小瓶,并且把风干的试剂条(纸面朝下)放入含蓝色液体的小瓶中,等10分钟,拿出试剂条并用自来水冲洗。

 

 

 

 

Step 6:第六步
用提供的色卡比较试剂条,任何颜色变化都表示有雪卡毒素的存在,颜色越深,毒素越多。

 

Cigua质控试验


所有生化检测都会由于时间、保存条件等变质。本试剂盒提供了一个一次性的质控试验,可以检查试剂条的灵敏度。如果试剂条放置了很久,在做实验之前应该先做这个质控试验。

质控试验:

一个阳性的质控试验表明试剂条仍然对毒素敏感。从一个红盖的小瓶中拿出质控试剂条。从第五步开始做,轻轻摇动含蓝色液体的小瓶,放入质控试剂条,等10分钟。用自来水冲洗,试剂条上的很淡的蓝色表明(0.8-1.0ppb),试剂条仍然敏感。

你应该再做一个阴性试验进一步验证。从第三步开始做(但不要放入鱼样品),这个试验应该没有任何颜色变化。

如果这两个实验有一个没有达标,并且试剂盒已在保质期外,表明试剂盒已经不能使用了。

Using a three step process, it provides a color coded result.  A blue results means ciguatera is present; a clear result means the fish is safe to eat.  Each test also includes a positive and negative control test to allow you to test for any shipping related damage to the test.

Ciguatera produces serious gastro-intestinal and neurological pathologies with no known cure.  Therefore, the best medicine if proper prevention.

Step 1:
While cleaning your fish, use the tweezers provided to remove a small, rice-grain-sized, sample of meat from the fish underneath the skin. Take care not to touch the sample with your fingers.
Step 2:
Put the fish sample into the bottom of the vial containing the clear liquid.  Caution: Oils from your hands may contaminate the test. 
Step 3:
Remove a Test Stick from the long tube and place it, paper-covered end down, into the vial containing the clear liquid and the fish sample. Wait 20 minutes.
Step 4:
Clean tweezers with water and return to holding slot in kit. Remove the Test Stick and allow it to thoroughly air dry for 15 to 20 minutes. Do not touch white paper-covered end of the stick with hands or fingers.
Step 5:
Gently shake vial with blue liquid to mix solution. Open and place the dry Test Stick (paper-covered side down) into the vial containing the blue liquid solution. Wait for 10 minutes, remove the Test Stick and gently rinse in tap water.
 
Step 6:
Compare the color on the end of the Test Stick to the color strip provided on the inside box lid. Any color change on the Test Stick indicates that the fish contains ciguatera poison and should not be eaten. The darker the color the more poison the fish contains.

 

Control Test:
All biochemical tests are subject to degradation over time depending upon storage conditions. A one-time positive control test is provided to check the sensitivity of the kit. This control test should be performed to verify the sensitivity of the test before testing any fish intended for consumption.

Positive Control Test:

A positive control test assures that the kit is still sensitive to toxin. Remove the Positive Test Stick from the Red-Capped vial. Begin instructions at Step 5. Gently shake vial containing blue liquid first. Place Test Stick in vial containing blue liquid and wait 10 minutes. Rinse in tap water. A very faint blue (positive) color on the stick (between 0.8 and 1.0 ppb) indicates that the kit is working properly.

You should also conduct a negative control test to further verify the test kit. Begin the instructions at Step 3 (don't use any fish tissue) using any of the paired test vials. This test should result in a Test Stick with no color.
