Actigel ALD Ultraflow 4 is an activated resin for immobilizing primary amines from small ligands to large proteins. Convenience and economy are the key advantages of Actigel ALD Ultraflow 4: coupling is simple, rapid and highly efficient.

During coupling, an instantaneous, reversible Schiff-base reaction takes place between the monoaldehyde groups and the primary amines of the ligand. The Schiff-base is then reduced to a stable secondary amine by the addition of a reducing agent so mild that it cannot reduce intramolecular disulfide bridges. ALD couples both acidic and basic proteins with the same high efficiency, typically >90% due to the high density of stable aldehyde groups.

Coupling is highly reproducible. Hydrolysis or inactivation of the activated groups is the main cause of irreproducible coupling, frequently encountered with traditional coupling chemistries.

Using ALD chemistry, coupling efficiency is simply a function of loading. No hydrolysis or inactivation of active groups takes place during the coupling reaction. This provides a highly reproducible coupling of ligands, critical for both research and manufacturing.
